Dr. McLean Bolton’s lab aims to characterize the morphology of neurons in the CNTNAP2 (contactin-associated protein-like 2) mouse model of Autism. The CNTNAP2 gene has been characterized to be responsible for cortical dysplasia and focal epilepsy (CDFE) syndrome. 70% of individuals affected by CDFE have Autism Spectrum Disorder (ASD). Distinct pathways lead to the social and repetitive deficits in ASD and CNTNAP2 mice are useful to test new pharmacological treatment. Analysis of CNTNAP2 mice’ neuronal morphology will show whether number or type of synaptic connections to the neuron is likely to differ or if the neuron will process information differently due to size and shape. Immunohistochemistry, a method of identifying discrete tissue components by the interaction of target antigens with specific antibodies tagged with fluorescents, will be utilized to see the neurons. Imaging will occur utilizing confocal microscopy. Image reconstruction software will be utilized to quantify the morphology.

I will be assisting a wildlife ecologist in monitoring a population of endangered woodpeckers in Picayune Strand State Forest. This will involve assisting with chick banding, roost monitoring, band identification, fledgling checks, and cavity maintenance. Specific tasks include carrying Swedish climbing ladders and other equipment to nesting sites and documenting the banding process, using spotting scopes to read leg bands, and conducting foraging follows to locate fledglings and documenting survivorship. The second component is a week in South Carolina that will involve vegetation monitoring of a restored pocosin wetland and photo point documentation of hydrologic conditions. This will involve walking belt transects to sample survivorship of planted shrubs and documenting the presence of hardwood wetland species in 13, 50 x 40 meter belt transects. I will be taking photographs at established stations throughout the wetland as well.

After undergoing a full faculty employment process, I was able to begin volunteering. I am trained in carrying out duties on the second floor, third floor, and at the front desk. This internship will require that I learn and understand the laboratory procedures and associated safety procedures of the synthesis that I will be working on. My first task will be to prepare approximately fifty milligrams of a novel anti-cancer compound that inhibits KLF5-mediated tumor growth, for use in workup and analysis that will determine whether or not the compound is of enough interest to synthesize a useful quantity for animal trials. I will be learning the theory behind the synthetic pathway already established to arrive at the target compound, as well as the laboratory procedures to prepare, monitor, isolate, and verify the identity and purity of the product. I will be responsible for the proper execution of these techniques and procedures on a larger scale once I have prepared the 50mg amount, aiming for perhaps .75g of the compound, and the possible synthesis of structural analogs of the compound.
